The similarities between FPLC and HPLC

The infusion pump, highly sensitive detector, gradient elution device, automatic collection device, computer control and other equipment are all used by FPLC and HPLC. FPLC and HPLC are also characterized by high speed, high resolution and high separation efficiency.

 

The differences between FPLC and HPLC

The pipeline system of FPLC is composed of high pressure resistant plastic pipes. The mobile phase is driven by a plunger pump, with medium and low pressure. The chromatographic column with a large inner diameter is used. The analytical accuracy is high. The flow rate range is 0 ~100 mL/min (Z large flow rate can reach 1000 mL/min). The analytical accuracy is lower than that of HPLC. Only conventional inorganic salt solution is used, which is very suitable for large-scale purification of samples.

Most of the materials used in the pipeline system of HPLC are metals. The reciprocating pump or diaphragm pump drives the mobile phase together. With high pressure, the chromatographic column with small inner diameter is used, which has high analysis accuracy. The flow rate Z is generally not higher than 20 mL/min (some preparation grade HPLC can reach 300 mL/min). Sometimes organic solvents with a certain concentration need to be used, which is applicable to the detection of micro samples or small amount of purification preparation.
